difficult issue with a 5.72 web connect site running on Win Server 2012 R2 64 bits ...
works fine (in file mode as COM mode brings up a flock of COM servers), until it once stops brutally:
- /js/mpn.js : response OK
- /admin/admin.aspx : no response - hourglass
- RDP on the server displays a black screen
- other sites on the same machine using another app pool respond OK
any idea?

To make sure that's the problem create a simple ASPX test page (test.aspx) and put:
<%= DateTime.Now %>
inside and make sure that works. if it does the problem you might be having with the admin page is authentication - make sure that Windows and Basic Auth are enabled on the site.
Not sure what would make it 'hang' - usually if ASP.NET is not installed you get a 4xx error of some sort.

If admin.aspx doesn't work then there's a problem with ASP.NET in that particular app pool. Make sure that ASP.NET is enabled with Integrated Pipeline (ASP.NET 4.x preferrably).
